Currently, different genes are known to have associations with diabetes, one of which is CYP3A4 gene on chromosome 7. This gene belongs the largest number of substrates regarding cytochrome p450 enzymes. CYP3A4 gene is found in endoplasmic reticulum of all tissues except brain; however, the main source of its protein accumulates in the liver and prostate. The main purpose of the present study is to investigate whether there is any predictable associations between specific variants of CYP3A4 gene and type II diabetes. Materials and Methods: In this case-control study, 100 blood samples of healthy individuals as well as 100 blood samples of patients referring to diabetes clinic of Ali Asghar in Zahedan were collected. DNA extraction was performed using phenol-chloroform method combined with proteinase K. The determination of individuals' genotype was done by a sequencing method. Results: The results of the study showed no significant differences (causing to alter protein conformation) between DNA samples of type two diabetic patients and those of healthy samples in terms of nucleotide sequence in CYP3A4 gene.
